The event is a round of British Cycling's National series.
The opening prologue time trial is over a 1.29km course from the Sea Terminal to the War Memorial and starts at 5.45pm.
Tomorrow sees road races at Peel and Ellerslie with the action moving to Ramsey on Sunday.
Leading the Junior challenge for the Isle of Man will be Matt Bostock, Leon Mazzone, Jamie Dudley and Joe Harris.
